Library reagents for whole exome sequencing

This public tender process is for reagents and kits used for the diagnostic whole exome sequencing (WES) NGS pipelines of HUS Diagnostic Center, Laboratory of Genetics. The pipeline is used for preparing high quality sample libraries for Illumina NovaSeq 6000 sequencer. The HUSLAB WES workflow is based on a target enrichment step using hybridization probes to enrich DNA fragments originating from the genomic Regions of Interest (RoI). HUSLAB WES is used for high sensitivity variant detection of both germline and somatic DNA variations.

HUSLAB WES process throughput is estimated to be 2000 samples in one year. The Contracting Entity estimates that orders will be made two (2) times per year, as separate orders so that separate order includes supplies needed for six (6) month use. One-time order will be the amount of reagent for processing of 900-1000 samples. One-time order may be larger or smaller, depending on actual sample numbers at a given time period.

Library preparation using as low as possible number of different reagent kits is preferred i.e. offering Kit 1 for one required item and a separate Kit 2 for another item is not preferred and if so offered, the total cost of both kits must be included in the price comparison although only a portion of kit components are used for the pipeline.

The following components are requested for the HUSLAB WES process

- Library preparation reagents to produce NovaSeq 6000 compatible paired-end libraries from genomic DNA.

- Capture and target enrichment reagents compatible with the adapters and probes already in use in the HUSLAB WES process (see section Procurement object criteria for details). Adapter blockers and SPRI cleanup beads are requested to be included in the offer.

INTENDED USE

The reagents offered will be Components of the HUSLAB WES process. HUS Diagnostic Center will select the Components and combine them in order to create the final HUSLAB WES process used for generation of high quality and high sensitivity customized WES sample libraries, fulfilling the Special Requirements for High Sensitivity Variant Detection.

The WES pipelines using the reagents are used for production of high quality WES sample libraries from DNA isolated from:

- whole blood

- freshly isolated or frozen tissue samples or cells

- FFPE samples (optionally omitting DNA fragmentation)

The NGS pipelines using the Components allow for HUS Diagnostic Center to build NGS pipelines fulfilling the accreditation requirements of SFS-EN ISO 15189:2012.

The offered products are subject to a quality comparison during the trial use. The contracting entity does not have the opportunity to carry out a laborious trial use for a group of tenderers larger than the stated.
